Richard Castle is a ridiculously rich and famous murder mystery novel author. Being rich and famous somehow enabled him to become friends with the mayor of New York City (the mayor isn't actually a character in the show, but Castle brings him up a lot). Richard got the "okay" from the mayor to "shadow" Kate Beckett, an NYPD homicide detective, for his upcoming book.
The entire show is just about all the murders that Kate has to investigate. Castle turns out to be a huge help, considering he is a murder mystery writer.
After Castle finishes his book, "Heat Wave," in which the main character is homicide detective Nikki Heat (clearly modeled off of Kate), he remains at Kate's side, basically a detective himself.
While the show does have its fair share of drama, I'd say its mostly comedy, because Castle is hilarious. He always has the perfect thing to say. The situation of Kate's mother being murdered (before the time frame of the show) adds the drama aspect that the show needs.
The last thing I need to add is that Beckett and Castle are in love, they just don't admit it to each other, or even themselves. It's bound to come out sometime though, right?
